Heterosphaeria-like on Prunus padus
Viktorie Halasu, 16-04-2018 11:39
Viktorie HalasuHello forum,

I have an interesting discomycete found on bark of (living?) Prunus padus. First I thought Heterosphaeria cf patella, but:
- medulla is not gelified
- paraphyses apex looks different, not so enlarged, no VBs
- there's a gel layer on receptacle surface
- different spore size, IKI+bb asci.
- excipulum turns dark brown-olive in KOH

Some characters remind me on Mollisia (KOH reaction, white hymenium when drying), but others on Heterosphaeria. Growing in lowland, former wetland forest (mostly Fraxinus and Populus spp.), Dermea padi and Biscogniauxia granmoi were found nearby. Apothecia 0,5-1 mm wide.
Thank you in advance for any advice.

Dear Viktorie

Great, I cannot remember such a fungus. Did you study the anamorph also? How sure does this belong to the apothecia in your opinion?

Dear Zotto,

thank you very much. I feared I'll get answer like this since I didn't find anything similar in your folder. : )

I didn't study it yet in detail, I'll try to do that today evening and test the KOH as well. But here's the upper part of one conidioma (pyknid? don't know correct term) where I measured the conidia and it has the same gel layer. I don't know how common this feature is among inoperculates.

I wanted to test it with CRB, but still have some problems with this reagent - either it's too concentrated (and staind everything violet) or I get no reaction at all. Could you please advise me the correct way to apply it? I tried it with the medium becoming only light blue, which avoided the overstaining, but never got those beautiful turquise VBs (in other species).

I simply add CRB to the waer mount and let it diffuse. The size of the dopr determines the concentration. Of course, CRB must be aqueous, not dissolved in another reagent.

Not sure what you want, maybe staining of the gel layer? That should work, usually.

Re : Heterosphaeria-like on Prunus padus
Bonjour,

Elisabeth

Re : Heterosphaeria-like on Prunus padus
Dear Elisabeth,

great, it's indeed the same fungus. Would you please send me the collection data? Meanwhile I identified it as Glutinium laevatum, a species described several times in anamorph stage.
edit: was it on Padus avium, or P. serotina?

If you ever find something similar on Salix (maybe shrub Salix?), we would be much interested in studying and sequencing it. (There's a sister species, perhaps less common, G. palinum. I have Petrak's locality nearby, but haven't found it there yet.)
