récolte faite en tourbière avec sphaignes, apothécies sur de petites tiges pourrissantes. Apothécies cupuliformes, non stipitées. Asques d'une moyenne de 105 µm, très faiblement amyloïdes. Spores 11-16 x 3,5-5 µm.
Merci d'avance pour vos suggestions.
Harvest made with sphagnum bog, apothecia on small rotting stems. Apothecia cup-shaped, without stipe. Asci 105 µm, très faiblement amyloïdes. Spores 11-16 x 3,5-5 µm.
Thank you in advance our suggestions.

Re : Asco blanc, Hymenoscyphus ???
Hi Patrice
is it actually MLZ what you used? This could be a Pezoloma, and a hemiamyloid reaction of the apical ring would be possible.
The externe layer could be the gelatinized outer layer typical of the genus.

Melzer 2015.
Nothing with Lugol.

Re : Asco blanc, Hymenoscyphus ???
Does this mean you used MLZ bought in 2015? And als only very pale blue reaction in Lugol?
If you don't have Lugol, try KOH-pretreatment whether the reaction gets more intense.

Re : Asco blanc, Hymenoscyphus ???
Sorry, I specify :
- MLZ bought in 2015, yes, and very very pale blue reaction, or no reaction.
- I have Lugol bought in 2014, and the reaction is negative.

Re : Asco blanc, Hymenoscyphus ???
This is unusal that MLZ gives a more distinct reaction. I see a pale blue on your pics. If your Lugol is still dark brown (it should so), and if you still have material, it would be helpful to see a mount in IKI.

Re : Asco blanc, Hymenoscyphus ???
I have no IKI, sorry.
I also tried :
- KOH then melzer : nothing better
- KOH then lugol : no reaction
My lugol is color amber.

Re : Asco blanc, Hymenoscyphus ???
Lugol = IKI, but the concentration is possibly too week when it is amber in the bottle. Look my homepage, last photo
http://www.invivoveritas.de/iodine-reaction/
anyway it shoudl give a reaction also when yellow-amber.

Re : Asco blanc, Hymenoscyphus ???
No doubt, my lugol is too clear, it is surprising because it is not old.

Re : Asco blanc, Hymenoscyphus ???
If you bozught it as Lugo it has usually about 0.3% iodine. This is the standard and useful for lichens, although I prefer also there a higher concentration because some blue reactions turn red only then.

Re : Asco blanc, Hymenoscyphus ???
Hello
Concentration of the Lugol provided to Patrice is 1 % iodine, 2% KI pour 100 ml.
I have now requested that KI raises to 3% for the Lugol reactive
Is it convenient for you?

Re : Asco blanc, Hymenoscyphus ???
Hi Francois
I put the KI and crystals without using any weighing machine, so I cannot say for sure that my concentration is 1%. When I wrote my article in 1987 I measured it exactly, and there noticed the decrease in concentration during some months if you keep it in small bottles with a plastic lid.
Maybe I presently use a better lid, it seems not to permit the iodine to escape. When did Patrice get the solution?
3% maybe o.k., but it is then more difficult to observe the blue part of the RB reaction and you easily get the preparation overstained.
