Malcolm  GreavesThis was found on a roadside bank on moorland. Am I right in thinking this could be Microglossum fuscorubens or M rufescens. According so some these are both just variants of M olivasceum.
What is the differences between these two?

Today you need a sequence :-)   But to come closer I would appreciate some more photos, lying horizontally, and a photo of the ascus apex in IKI, also to verify the croziers.

This is great! The iodine reaction is in MLZ, KOH-pretreated?
The ascus base looks like simple-septate with protuberances, a strange feature in Microglossum.

Hlepful would be a scope in spore size.

I downloaded your images hoping that the sample gets a sequence sometime.

If you don't mind I would add the date, locality and vegetation type.

Spores were (12.9)14.35 - 15.3(16.5) x 4.04-4.98 (av 10 spores 14.78 x 4.45)
It was found 31/10/17 in moorland at Levisham 54.18 N 0.44 W.
I am sending sample for DNA work.

This is good, thanks. You can see that the stained spores are distinctly narrower than the others. What medium is it? And in which montant were the measures taken?

Is the stained ascus apex KOH-pretreated? (becaus eof possible reddish stains in Lugol).

The top two photos on the left were in Meltzers the one with droplets in water and these were from a squash. The others, stained with Plaqsearch were from a spore drop so that might account for the spore size. The measurements were from the spore drop. Checking the others the length is still the same but they were up to 5.5 wide.
The asci tops were directly mounted in Meltzers no KOH or heat treatment. I will try with Lugol and a pretreatment of KOH and see if I get any different results.

A spore drop is fro shot spores? They should be mature and not smaller. What is Plaqsearch, I never heard? Seems you are a dentist :-) It is strange that the spores died in that mountant, how can it then be applied to a patient? Or did you keep the spore print for some time in the dry state? In any case I wonder why the Melzer's spores are wider (Melzer kills the spores).

It will be enough to check the Lugol reaction. it could be that high concentration turns it dirty red. But the Lugol must have more than 0.3% iodine for this test (must look deep redbrown).

The spore "drop" was a fruitbody left on a slide overnight.
Plaqsearch is a water based stain primarily for teeth but it contains Bryliant blue and phloxine which are often absorbed differently and can show good micro detail.
I have tried the asci with Lugol (and Baral's which looked darker) both giving a blue reaction.

In case you have left the spores for a longer time in such a dye they will die. Basic dyes enable vital staining but only for some miniutes or perhaps a quarter of an hour.
